Features of the water regime and photosynthesis in hybrid forms of cherry plum under the dry wind season conditions

Background. The results of a study on the drought resistance of cherry plum hybrid forms are presented. Water retention capacity of leaves and chlorophyll fluorescence were measured under the simulated conditions close to the dry wind season.Materials and methods. The water regime parameters were studied for five cherry plum genotypes in 2021 and 2022. Leaves of four hybrid forms developed at the Nikita Botanical Gardens and the reference cherry plum hybrid cultivar ‘Rumyanaya Zorka’ (Prunus rossica Erem.) were taken into the experiments. Using the guidelines by G. N. Eremeev and A. I. Lishchuk, a series of experiments on controlled leaf wilt at the air temperature of +27°C and relative humidity of 30% were conducted on cherry plum to assess the functional state of the photosynthetic apparatus during the development of water stress under dry wind conditions. The experiments were carried out in a Binder climate chamber. Chlorophyll fluorescence was measured using a portable Floratest fluorometer. The measurements were based on the parameters of the chlorophyll fluorescence induction curve (CFI) obtained on native leaves. The leaves of cv. ‘Rumyanaya Zorka’ in the state of complete watering served as the reference.Results and discussion. Dependence was identified between chlorophyll fluorescence and moisture content in the leaves of the studied samples. The hybrids were found to respond to drought in different ways. A relatively increased ability to retain moisture during dehydration was observed in form 12/26 whose leaves gave off moisture more slowly and restored their turgor better. It is promising for cultivation in areas with insufficient water supply. Form 99/36 had the lowest potential drought resistance. Forms 99/14 and 9/28 occupied an intermediate position.

Effects of Cu2O nanoparticles on the kinetic characteristics of rapid chlorophyll fluorescence induction and related genes in wheat seedlings.

Metal nanoparticles could be accumulated in soils, which threatens the ecological stability of crops. Investigating the effects of cuprous oxide nanoparticles (Cu2O-NPs) on photosystem Ⅱ (PSⅡ) of wheat seedling leaves holds considerable importance in comprehending the implications of Cu2O-NPs on crop photosynthesis. Following the hydroponic method, we investigated the effects of 0, 10, 50, 100, and 200 mg·L-1 Cu2O-NPs on chlorophyll fluorescence induction kinetics and photosynthetic-related genes in wheat seedlings of "Zhoumai 18". The results showed that, with the increases of Cu2O-NPs concentrations, chlorophyll contents in wheat leaves decreased, and the standardization of the OJIP curve showed a clearly K-phase (ΔK＞0). Cu2O-NPs stress increased the parameters of active PSⅡ reaction centers, including the absorption flux per active RC (ABS/RC), the trapping flux per active RC (TRo/RC), the electron transport flux per active RC (ETo/RC), and the dissipation flux per active RC (DIo/RC). Cu2O-NPs stress decreased the parameters of PSⅡ energy distribution ratio including the maximum quantum yield of PSⅡ (φPo), the quantum yield of electron transport from QA (φEo), and the probability that a trapped exciton moved an electron further than QA (Ψo), while increased the quantum ratio for heat dissipation (φDo). Moreover, there was a decrease in photosynthetic quantum yield Y(Ⅱ), photochemical quenching coefficient (qP), net photosynthetic rate (Pn), stomatal conductance (gs), intercellular CO2 concentration (Ci), and transpiration rate (Tr) of leaves with the increases of Cu2O-NPs concentration. Under Cu2O-NPs stress, the expression levels of genes which included PSⅡ genes (PsbD, PsbP, Lhcb1), Rubisco large subunit genes (RbcL), cytochrome b6/f complex genes (PetD, Rieske), and ATP synthase genes (AtpA, AtpB, AtpE, AtpI) were downregulated. These results indicated that Cu2O-NPs stress altered the activity and structure of PSⅡ in wheat seedlings, affected the activity of PSⅡ reaction centers, performance parameters of PSⅡ donor and acceptor sides. PSⅡ related genes were downregulated and exhibited significant concentration effects.

Application of Chlorophyll Fluorescence Analysis Technique in Studying the Response of Lettuce (Lactuca sativa L.) to Cadmium Stress.

To reveal the impact of cadmium stress on the physiological mechanism of lettuce, simultaneous determination and correlation analyses of chlorophyll content and photosynthetic function were conducted using lettuce seedlings as the research subject. The changes in relative chlorophyll content, rapid chlorophyll fluorescence induction kinetics curve, and related chlorophyll fluorescence parameters of lettuce seedling leaves under cadmium stress were detected and analyzed. Furthermore, a model for estimating relative chlorophyll content was established. The results showed that cadmium stress at 1 mg/kg and 5 mg/kg had a promoting effect on the relative chlorophyll content, while cadmium stress at 10 mg/kg and 20 mg/kg had an inhibitory effect on the relative chlorophyll content. Moreover, with the extension of time, the inhibitory effect became more pronounced. Cadmium stress affects both the donor and acceptor sides of photosystem II in lettuce seedling leaves, damaging the electron transfer chain and reducing energy transfer in the photosynthetic system. It also inhibits water photolysis and decreases electron transfer efficiency, leading to a decline in photosynthesis. However, lettuce seedling leaves can mitigate photosystem II damage caused by cadmium stress through increased thermal dissipation. The model established based on the energy captured by a reaction center for electron transfer can effectively estimate the relative chlorophyll content of leaves. This study demonstrates that chlorophyll fluorescence techniques have great potential in elucidating the physiological mechanism of cadmium stress in lettuce, as well as in achieving synchronized determination and correlation analyses of chlorophyll content and photosynthetic function.

The Fitting of the OJ Phase of Chlorophyll Fluorescence Induction Based on an Analytical Solution and Its Application in Urban Heat Island Research

Chlorophyll (Chl) fluorescence induction (FI) upon a dark–light transition has been widely analyzed to derive information on initial events of energy conversion and electron transfer in photosystem II (PSII). However, currently, there is no analytical solution to the differential equation of QA reduction kinetics, raising a doubt about the fitting of FI by numerical iteration solution. We derived an analytical solution to fit the OJ phase of FI, thereby yielding estimates of three parameters: the functional absorption cross-section of PSII (σPSII), a probability parameter that describes the connectivity among PSII complexes (p), and the rate coefficient for QA− oxidation (kox). We found that σPSII, p, and kox exhibited dynamic changes during the transition from O to J. We postulated that in high excitation light, some other energy dissipation pathways may vastly outcompete against excitation energy transfer from a closed PSII trap to an open PSII, thereby giving the impression that connectivity seemingly does not exist. We also conducted a case study on the urban heat island effect on the heat stability of PSII using our method and showed that higher-temperature-acclimated leaves had a greater σPSII, lower kox, and a tendency of lower p towards more shade-type characteristics.

Psammophytes Alyssum desertorum Stapf and Secale sylvestre Host Are Sensitive to Soil Flooding

While morphological and functional traits enable hydrophytes to survive under waterlogging and partial or complete submergence, the data on responses of psammophytes—sand plants—to flooding are very limited. We analyzed the effect of 5- and 10-day soil flooding on the photosynthetic apparatus and the synthesis of alcohol dehydrogenase (ADH), heat shock proteins 70 (HSP70), and ethylene in seedlings of psammophytes Alyssum desertorum and Secale sylvestre using electron microscopy, chlorophyll a fluorescence induction, and biochemical methods. It was found that seedlings growing under soil flooding differed from those growing in stationary conditions with such traits as chloroplast ultrastructure, pigment content, chlorophyll fluorescence induction, and the dynamics of ADH, HSP, and ethylene synthesis. Although flooding caused no apparent damage to the photosynthetic apparatus in all the variants, a significant decrease in total photosynthesis efficiency was observed in both studied plants, as indicated by decreased values of φR0 and PIABS,total. More noticeable upregulation of ADH in S. sylvestre, as well as increasing HSP70 level and more intensive ethylene emission in A. desertorum, indicate species-specific differences in these traits in response to short-term soil flooding. Meanwhile, the absence of systemic anaerobic metabolic adaptation to prolonged hypoxia causes plant death.

The influence of powdery mildew (Sawadaea bicornis (Wallr.) Miyabe) on chlorophyll fluorescence in Norway maple (Acer platanoides L.) seedlings

The article presents the results of research on impact of Sawadaea bicornis (Wallr.) Miyabe on the state of photosynthetic apparatus in Acer platanoides L. seedlings using a technique of chlorophyll fluorescence induction (ChlF) measurement, which at the present time can be implemented through the use of biosensors. The research was conducted in September 2022 in the territory of the Botanical Garden of Oles Honchar Dnipro National University. To diagnose a violation of the native chlorophyll photosynthesis in fresh leaves of A. platanoides, a portable fluorometer “Floratest” was used (selected spectral range for fluorescence intensity measurement was 670–800 nm). The research was carried out on fresh leaves of Norway maple seedlings both not unaffected and affected with powdery mildew. Analysis of the data obtained indicates a high sensitivity of the parameters of chlorophyll fluorescence induction to damage by the disease regardless of environmental conditions of local growth of A. platanoides seedlings. The high informativeness of induction changes in chlorophyll fluorescence in the structural organization of chloroplasts in Norway maple leaves determined by the parameters Fo, Fm, Fv, Fv/Fо, Fv/Fm, (Fm–Fst)/Fst, (Fp–Fо)/Fv was revealed. This study demonstrated the effectiveness of using the studied chlorophyll fluorescence parameters to detect severe stress in Norway maple seedlings caused by powdery mildew exposure when the fungus affects more than 50% of the leaf blade area. It is further necessary to conduct dynamic studies throughout the growing season to determine the effectiveness of using these parameters to detect mild stress in the early stages of infection. Therefore, our research showed that the method of induction of chlorophyll fluorescence using the “Floratest” device allows you to quickly determine the general condition of the plant by evaluating photosynthesis, which is the main life process. Analysis of chlorophyll fluorescence parameters is a powerful and effective tool for determining the effect of a parasitic fungus on a plant.

Exogenous Melatonin Alleviates the Inhibitory Effect of NaHCO3 on Tomato Growth by Regulating the Root pH Value and Promoting Plant Photosynthesis

Soil salinity is a severe threat to agricultural production. Most saline soils turn alkaline, increasing the soil pH and, in turn, hampering the growth and development of crops. In this study, the effects of a foliar spray of melatonin (MT; 100 μmol·L−1) on the pH of the root environment, growth of tomato seedlings, endogenous MT levels, rapid chlorophyll fluorescence induction kinetics, and key enzymes of the Calvin cycle under alkaline (60 mmol·L−1 NaHCO3) stress were studied in Riegel 87-5 tomatoes. The results revealed that the growth and photosynthesis of tomato seedlings were inhibited by increased pH in the root environment under alkali stress; however, the application of exogenous MT reduced the pH of the root environment, alleviated the inhibition of growth of tomato seedlings under alkali stress, increased the content of photosynthetic pigments, alleviated the damage of the donor and acceptor sides of the photosynthetic electron transport chain, increased the activity and efficiency of photosynthetic electron transport, and optimized the share of the light energy allocated to PSII reaction centers. Increased expression levels of Calvin-cycle enzymes, including fructose-1,6-bisphosphate aldolase (FBA), fructose-1,6-bisphosphate esterase (FBP), and phosphoglycerate kinase (PGK), led to enhanced photosynthetic performance in tomato seedlings. Exogenous MT boosted endogenous MT levels and stimulated the production and secretion of organic acids in the root system. This regulation of organic acid content reduced the environmental pH in the inter-root zone, alleviating the damage caused by alkali stress. This study indicated that the exogenous administration of MT may mediate an increase in endogenous MT levels, regulate the efficiency of photosynthesis and root pH levels, and play a crucial role in mitigating injury caused by alkali stress in tomato seedlings.

Effects of Elevated CO2 Concentration and Temperature on the Growth and Photosynthetic Characteristics of Populus simonii × P. nigra ‘1307’ Leaves

This study aimed to establish the effects of elevated CO2 concentration and temperature on the photosystem II (PSII) performance and photosynthetic characteristics of Populus simonii × P. nigra ‘1307’ leaves. Different CO2 concentrations (400 and 800 µmol·mol−1) and temperatures (room temperature and room temperature +2 °C) were set in artificial climate change simulation and control chambers, and the rapid chlorophyll fluorescence induction kinetics curve (OJIP curve) of Populus simonii × P. nigra ‘1307’ was determined. The generated OJIP curve was used to analyze the change characteristics in photosynthetic performance. The results revealed that under elevated temperature conditions, the concentrations of chlorophyll a, b in Populus simonii × P. nigra ‘1307’ leaves were significantly increased. At the same time, there were no significant changes in the chlorophyll concentration under the superimposed effect of elevated CO2 concentration and temperature. The PSII comprehensive performance index (PIABS) of Populus simonii × P. nigra ‘1307’ was significantly inhibited under elevated temperatures due to the increased closure degree (Vj) of the PSII reaction center and the damage of the receptor side. This reduced the electron transfer capacity per unit reaction center (ETo/RC) and unit cross-sectional area, which decreased the quantum yield of the electron transfer. Under the elevated CO2 concentration, ETo/RC was also inhibited. Still, PIABS was enhanced owing to the increased number of active PSII per unit area and the low reduction rate of the primary quinone receptor (QA). Under the superimposed effect of the two factors, the electron transfer performance of the donor and receptor sides of PSII was improved compared to the treatments only subjected to elevated temperature; thus, PIABS was not significantly reduced compared to the control. Therefore, the continuous increase in temperature by 2 °C significantly inhibits the electron transfer capacity of the photosynthetic system of Populus simonii × P. nigra ‘1307’ leaves. On the other hand, an increase in CO2 concentration expands the PSII reaction center, while enhancing the electron transfer capacity of the donor and receptor sides, which alleviates the photosynthetic inhibition caused by the elevated temperature.

Effects of UV radiation on photosynthesis of Sargassum muticum

As an invasive species of macroalgae, Sargassum muticum (Yendo) Fensholt has invaded European shores and the Mediterranean, and even drifting individuals have been observed in sea area near the Canary Islands, becoming a potential species responsible for the “Sargassum golden tide”. When drifting on the sea surface, algae receive more ultraviolet radiation (UVR). However, photosynthetic responses and adaptation mechanism of S. muticum to UVR remain unclear. In this study, thalli of S. muticum were respectively exposed under two radiation treatments of 200 W m−2 PAR (photosynthetically active radiation, 400–700 nm), and 200 W m−2 PAR + 38 W m−2 UVR (280–700 nm) for 120 mins, and then recovered at a low light (PAR) of 10 μmol photons m−2 s−1 for 240 mins. During this treatment, the photosynthetic properties of algae were measured, to investigate the responses and protection mechanism of S. muticum to UVR. The results showed that photosynthesis of thalli was significantly inhibited by UVR, reflected by decreased maximum photochemical quantum yield (Fv/Fm) and chlorophyll content, and changes of parameters of rapid light response curve and chlorophyll fluorescence induction curve in PAR and PAR + UVR treatments. Meanwhile, algae actively adopted several strategies to alleviate the inhibition caused by UVR, including photoprotection characterized by increased contents of UV-absorbing compounds (UVACs) and carotenoids (Car), excess light energy dissipation by elevated non-photochemical quenching (NPQ) coupled to the xanthophyll cycle, and photorepair manifested as an accelerated synthesis of D1 protein and enhanced antioxidant for scavenging ROS. Benefiting from the effects of the above several pathways, after 240 mins of low light recovery, photosynthesis was greatly restored in these two radiation treatments, with the restoration of Fv/Fm and parameters of rapid light response curve and chlorophyll fluorescence induction curve. Based on these results, we conservatively speculate that the photosynthesis of drifting S. muticum individuals may be restored in the evening with low light levels after being damaged by UVR on the surface of sea at midday, resulting in maintaining rapid growth and forming a golden tide.

Dependency of chlorophyll fluorescence of in vitro plant of Carlina L. on light conditions during their cultivation

Aim. To investigate the peculiarities of the functioning of the photosynthetic apparatus of plants of Carlina L. species under different light conditions of in vitro cultivation and under natural conditions using the method of induction of fluorescence of chlorophyll a. Methods. Methods of in vitro plants cultivation, a method of inducing fluorescence of chlorophyll a in light-adapted leaves. Results. It has been demonstrated that in vitro conditions reflect the evolved light growth requirements of species. Our previously obtained results regarding a closer match of physiological needs of in vitro plants of the species of Carlina onopordifolia Besser ex Szafer, Kulcz. et Pawt. to light conditions of variant 1.1 have been confirmed. Based on the synthesis of the dynamics of photosynthetic pigment content and their ratios in in vitro plants of Carlina acaulis L., as well as key parameters of fluorescence under different cultivation light conditions, a conclusion has been drawn about the greater alignment of variant 1.1 with the needs of plants of this species. It is hypothesized that in natural conditions, plants of the species of C. onopordifolia and Carlina cirsioides Klokov undergo abiotic stresses, manifested in increased dissipation of light energy into heat and the processes of photo-inhibition. There is significant enhancement in electron transport within the light-harvesting complex and reduction in the efficiency of quantum yield and light energy storage by Photosystem II. The vitality index of these species in their natural growth conditions is found to be half of the commonly accepted optimum value and 1.97–2.96 times lower compared to experimental in vitro plant groups. Conclusions. The obtained results suggest the usefulness of using the chlorophyll fluorescence induction method for assessing the functioning of the photosynthetic apparatus of in vitro plants of the genus Carlina.

Smart-Systems for Precision Agriculture, Environmental Protection and Healthcare

Appearance of innovative tools of informatics in the world is determined by development of information-communication technology, microelectronics, sensor and biosensor technologies. Spreading of these technologies on precision agriculture, environmental protection and health care gives opportunity to create smart fields, gardens, greenhouses, forests and parks, and also smart health monitors, which estimate the health state of person as in rehabilitation, so in emergency situations. Results of research of authors in the development of new information technologies and creation of main components of the smart systems for different purposes on this base are shows in this paper. Main requirements for knowledge bank for precision agriculture and ecological monitoring are defined. Main principles of creating knowledge bank on base of requirements are proposed. The typical smart system models and their development stages are considered. Structure of wireless multilevel networks for the estimation of state of biological objects of different origin and their nodes are described. Developed sensor networks and smart systems for agriculture are considered. Structure of proposed smart system for agriculture and ecological monitoring is given. Approaches for chlorophyll fluorescence induction curves analysis are studied. Results of network testing for the estimation of the autonomous work time of network nodes and possible errors are given. Wireless sensor network and smart system for remote medical monitoring are described. Diagnostic smart systems for estimation of quality of life are considered. Medical communicator, computer device on base of tablet computer, was used for their development. The short form of Survey Instrument (SF-36) for life quality estimation and abnormal uterine bleeding questionnaire, which was developed in State Scientific Institution "Center for Innovative Medical Technologies of the National Academy of Sciences of Ukraine", were embedded in communicator. STEPS questionnaire which was designed for epidemiological monitoring of the prevalence of noncommunicable diseases and their risk factors in the target subpopulations could be added to communicator. Development of smart systems for estimation of quality of life and prevalence of noncommunicable diseases was made in cooperation with R.E. Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology NAS of Ukraine (department of research management and innovation), Chebotarev Institute of Gerontology of the National Academy of Medical Sciences of Ukraine, Center for Innovative Medical Technologies of the National Academy of Sciences of the Ukraine. Canopy structural changes explain reductions in canopy-level solar induced chlorophyll fluorescence in Prunus yedoensis seedlings under a drought stress condition

Drought events have a major impact on vegetation structure and function. Recently, solar-induced chlorophyll fluorescence (SIF) has been widely used to understand the photosynthesis rates of vegetation under drought stress conditions. However, it is still unclear whether the reduction in SIF shown under drought stress conditions is regulated by physiological or structural factors. To understand the underlying reduction mechanism of SIF under drought stress, we conducted an experiment under a drought condition using one-year-old Prunus yedoensis seedlings. We compared the experiment trees with control trees that were not exposed to drought stress. We collected spectral and gas exchange data to monitor physiological changes and scanned the trees with a terrestrial laser scanner to monitor the structural changes. The discrete anisotropic radiative transfer (DART) and Fluspect models were used to simulate canopy-level SIF. We found that drought stress caused leaf-level steady-state fluorescence yield to increase, while maximum photosynthetic rate, stomatal conductance, and the quantum yield of Photosystem II decreased significantly. Regarding the canopy structural changes, the leaf inclination angle distribution of the experiment trees gradually turned toward erectophile over time (55.2 ± 9.3° to 74.7 ± 6.0°; mean ± standard deviation), whereas that of the control trees remained relatively constant (52.9 ± 8.8°). Furthermore, reduction of crown cover of the experiment trees was 3-fold (77.4 ± 9.8%) compared with the control trees (26.0 ± 16.2%). The simulated nadir-view canopy-level SIF of the experiment trees was reduced 2.8-fold compared to the control trees. These findings, obtained specifically from Prunus yedoensis seedlings, indicate that canopy-level SIF reduced due to the canopy structural changes, although leaf-level fluorescence yield increased. Therefore, canopy structural changes should be considered when attempting to understand SIF reduction in drought conditions.

Climate change is enforcing physiological changes in Arctic Ecosystems

Remote sensing and site-level observations have shown unprecedented changes in Arctic ecosystems owing to climate warming. These observations include greening and browning trends in Arctic vegetation as well as changes in species composition both in the tundra and the boreal forests. Here, we leveraged solar induced chlorophyll fluorescence (SIF) to study changes in ecosystem phenology across the pan-Arctic domain from 2000 to 2020. Ecoregions, as a proxy for plants’ functional traits, were the single most important variable to explain the spatial and phenological heterogeneity in observed SIF trends. We observed unique regional trends in ecosystems responses to climate change affecting the timing of spring photosynthesis onset, magnitude of peak productivity during the growing season (GS) and fall senescence. While, Photosynthetic activity in the early GS showed increasing trends across the vast majority of the pan-Arctic, it tends to decline at the end of the season for nearly half of the land area, including parts of North America but more significantly in central Siberia. The observed changes in phenology highlight the role of biodiversity in regional climate sensitivity and emphasizes the need for better representations of sub-biomes to community level information in Arctic ecosystem process models and projections. These results also highlight the importance of phenology information in ecosystem models for better understanding of the timing and magnitude of carbon uptake in the Arctic domain with accelerated changes in climate.

Variation in thermotolerance of photosystem II energy trapping, intersystem electron transport, and photosystem I electron acceptor reduction for diverse cotton genotypes

Cotton breeding programs have focused on agronomically-desirable traits. Without targeted selection for tolerance to high temperature extremes, cotton will likely be more vulnerable to environment-induced yield loss. Recently-developed methods that couple chlorophyll fluorescence induction measurements with temperature response experiments could be used to identify genotypic variation in photosynthetic thermotolerance of specific photosynthetic processes for field-grown plants. It was hypothesized that diverse cotton genotypes would differ significantly in photosynthetic thermotolerance, specific thylakoid processes would exhibit differential sensitivities to high temperature, and that the most heat tolerant process would exhibit substantial genotypic variation in thermotolerance plasticity. A two-year field experiment was conducted at Tifton and Athens, Georgia, USA. Experiments included 10 genotypes in 2020 and 11 in 2021. Photosynthetic thermotolerance for field-collected leaf samples was assessed by determining the high temperature threshold resulting in a 15% decline in photosynthetic efficiency (T15) for energy trapping by photosystem II (ΦPo), intersystem electron transport (ΦEo), and photosystem I end electron acceptor reduction (ΦRo). Significant genotypic variation in photosynthetic thermotolerance was observed, but the response was dependent on location and photosynthetic parameter assessed. ΦEo was substantially more heat sensitive than ΦPo or ΦRo. Significant genotypic variation in thermotolerance plasticity of ΦEo was also observed. Identifying the weakest link in photosynthetic tolerance to high temperature will facilitate future selection efforts by focusing on the most heat-susceptible processes. Given the genotypic differences in environmental plasticity observed here, future research should evaluate genotypic variation in acclimation potential in controlled environments.

A Sensitive Response Index Selection for Rapid Assessment of Heavy Metals Toxicity to the Photosynthesis of Chlorella pyrenoidosa Based on Rapid Chlorophyll Fluorescence Induction Kinetics.

Heavy metals as toxic pollutants have important impacts on the photosynthesis of microalgae, thus seriously threatening the normal material circulation and energy flow of the aquatic ecosystem. In order to rapidly and sensitively detect the toxicity of heavy metals to microalgal photosynthesis, in this study, the effects of four typical toxic heavy metals, chromium (Cr(VI)), cadmium (Cd), mercury (Hg), and copper (Cu), on nine photosynthetic fluorescence parameters (φPo, ΨEo, φEo, δRo, ΨRo, φRo, FV/FO, PIABS, and Sm) derived from the chlorophyll fluorescence rise kinetics (OJIP) curve of microalga Chlorella pyrenoidosa, were investigated based on the chlorophyll fluorescence induction kinetics technique. By analyzing the change trends of each parameter with the concentrations of the four heavy metals, we found that compared with other parameters, φPo (maximum photochemical quantum yield of photosystem II), FV/FO (photochemical parameter of photosystem II), PIABS (photosynthetic performance index), and Sm (normalized area of the OJIP curve) demonstrated the same monotonic change characteristics with an increase in concentration of each heavy metal, indicating that these four parameters could be used as response indexes to quantitatively detect the toxicity of heavy metals. By further comparing the response performances of φPo, FV/FO, PIABS, and Sm to Cr(VI), Cd, Hg, and Cu, the results indicated that whether it was analyzed from the lowest observed effect concentration (LOEC), the influence degree by equal concentration of heavy metal, the 10% effective concentration (EC10), or the median effective concentration (EC50), the response sensitivities of PIABS to each heavy metal were all significantly superior to those of φRo, FV/FO, and Sm. Thus, PIABS was the most suitable response index for sensitive detection of heavy metals toxicity. Using PIABS as a response index to compare the toxicity of Cr(VI), Cd, Hg, and Cu to C. pyrenoidosa photosynthesis within 4 h by EC50 values, the results indicated that Hg was the most toxic, while Cr(VI) toxicity was the lowest. This study provides a sensitive response index for rapidly detecting the toxicity of heavy metals to microalgae based on the chlorophyll fluorescence induction kinetics technique.

Effect of antigibberellins on morphogenesis, photosynthetic apparatus, productivity and their residual content in tomato fruits

The application of growth and development regulators on crops in order to optimize their production process is one of the leading tasks of modern plant physiology. Retardants – gibberellin inhibitors are widely used for this purpose. We investigated the effect of foliar treatment with EW-250, ССС-750 and 2-СEPA on morphogenesis, leaf apparatus, content of photosynthetic pigments, indices of chlorophyll fluorescence induction, CO2 gas exchange, and residual amounts of drugs in the fruits of tomato (Lycopersicon esculentum Mill.) Bobcat hybrid. The field experiment was laid on plots with an area of 33 m2. The treatment of the plants was carried out at the budding stage. Morphometric indices were determined at the stages of flowering and fruit formation. The chlorophylls content was determined in the raw material by the spectrophotometric method. Indices of photosystem II (PSII) photochemical activity were determined according to the parameters of chlorophyll fluorescence induction after a half-hour exposure of plants in the dark using a portable single-beam fluorimeter "Floratest". The determination of the residual content of retardants in the fruits was carried out on a Shimadzu GC gas chromatograph with a mass spectrometric detector – GCMS- QP2020 EI. All gibberellin inhibitors reduced linear plant size. The number of leaves on the plants decreased under 2-CEPA treatment, and increased after the application of EW-250. Treatment with 2-CEPA decreased, EW-250 significantly increased, and CCC-750 practically did not change the leaves’ fresh and dry weight. Leaf area and leaf index decreased under 2-CEPA treatment, but practically did not change when EW-250 and ССС-750 were applied. All antigibberellin drugs increased the leaf specific leaf weight and thickened the leaf lamina due to the growth of chlorenchyma cells. At the same time, growth inhibitors increased the volume of columnar parenchyma cells and practically did not change the size of spongy parenchyma cells. Retardants increased the chlorophylls content in leaves, while the ethylene producer 2-CEPA did not change this index. The plants’ chlorophyll index after treatment with drugs increased significantly. The whole plant dry weight increased under EW-250 treatment, decreased after 2-CEPA application, and did not change under CCC-750. It was established that the photosynthetic rate increased under the EW-250 treatment, both in the flowering stage and in the stage of fruit formation, while when using 2-CEPA and CCC-750, it occurred only at the stage of fruit formation. The most significant positive changes of PSII photochemical activity indices were observed under the use of EW-250. Under the action of the drug, the maximum and actual quantum efficiency of PSII increased, the linear electron transport accelerated, and the fraction of reaction centers that did not transfer electrons from the primary acceptor QA to QB decreased, at the same time the chlorophyll fluorescence decay coefficient significantly increased, which indicates an increase in the CO2 assimilation intensity. Retardants increased the proportion of the fruit in the whole plant dry weight. All growth regulators increased net photosynthetic efficiency. A significant increase in fruit yield occurred under EW-250 treatment. When using CCC-750, the index tended to increase, while under the influence of 2-CEPA the yield decreased. The residual amounts of EW-250 and CCC-750 in the fruits did not exceed the maximum permissible concentrations.

Adaptation of gooseberry varieties to the changed agro-climatic conditions of Kyiv Polissia

In connection with global warming, the study of plant adaptation to climate change, resistance to lack of moisture and high temperatures becomes relevant. Meteorological and phenological observations prove that a change in temperature conditions leads to an earlier start of vegetation and a lengthening of the growing season in all investigated gooseberry varieties. To assess the functional state of the photosynthetic apparatus of plants, a minimally invasive instrumental method of determining the intensity of chlorophyll fluorescence in plant leaves, adapted to work on the domestic photofluorimeter "Floratest", was used. Based on the results of the analysis of the parameters of the chlorophyll fluorescence induction curves (Fo, Fpl, Fmax, Ft) and the comparison of the coefficients for these values (Kpl, K1, K2), it was established that Fo and Kpl are the most informative. A strong correlation dependence of the indicator of the background level of fluorescence at the time of full opening of the shutter (Fo) was established. The value of Fо correlates positively with indicators of the moisture regime, negatively – with temperature indicators. Correlations between the fluorescence growth index and the variable (Kpl) showed the opposite: the largest positive correlation was observed with mean maximum temperature, the smallest with precipitation. On the basis of mathematical analysis and modeling of the obtained FIC curves and indicators of agro-climatic conditions, the values of the maximum temperature and the minimum amount of precipitation, which are critical for the functioning of the gooseberry photosynthetic apparatus, were determined. Such a study makes it possible to evaluate the course of photochemical reactions associated with the work of plant photosystems according to standard coefficients of photochemical activity, which have already been tested in numerous works of domestic researchers on a wide range of agricultural crops.

Physiological and Biochemical Changes in Soybean Plants Caused by Iodine-Selenium Chelates and Phytopathogenic Bacteria

The search for effective and environmentally sound measures to fight against plant diseases caused by phytopathogenic microorganisms is of great importance. It is especially important to investigate alternative measures to protect cultivated plants that combine elements beneficial to human health such as iodine and selenium. Purpose. The study of physiological and biochemical changes in soybean leaves after artificial inoculation of plants with different strains of phytopathogenic bacteria on the background of pre-sowing treatment of seeds with a solution of iodine-selenium (I-Se) chelates. Methods. Soybean plants of the Artemis variety were grown in field conditions. Before sowing, the seeds were treated with a 1% I-Se chelated solution (I — 80 mg/L and Se citrate — 0.05 mg/L). The experimental plants were inoculated with phytopathogenic bacteria from the microbiological collection of the Zabolotny Institute of Microbiology and Virology of the National Academy of Sciences, namely Pseudomonas savastanoi pv. glycinea IMВ B-9190, P. agglomerans IMВ B-9185, and P. syringae pv. syringae IMВ B-8531. The contents of chlorophyll-a, b and carotenoids in the leaves were determined by extraction in DMSO followed by spectrophotometry. Catalase activity was determined by the method of titrimetric permanganatometry with a 0.01M solution of KMnO4, the activity to non-specific peroxidases — according to Boyarkin’s method. Evaluation of the photochemical activity of photosynthesis according to the parameters F0, Fv/Fm, and RFd was carried out by the method of induction of chlorophyll fluorescence using a portable device «Floratest». Statistical processing of experimental data was carried out using the built-in functions of the Microsoft Excel program. Results. The increase of peroxidase activity of leaves was revealed after both pre-sowing treatment with 1% I-Se solution of intact plants and inoculation of them with different strains of bacterial pathogens in the following order: I-Se > I-Se+P. syringae pv. syringae 8531 > I-Se+P. agglomerans 9185 > I-Se+P. savastanoi pv. glycinea 9190. The catalase activity of leaves tissues increased only when infected with a specific pathogen P. savastanoi pv. glycinea 9190 (by 20.6%). After artificial inoculation with strains of both specific and facultative bacterial pathogens and the pre-sowing treatment with I-Se, there was observed an increase in the quantum efficiency of PSII (Fv /Fm) and fluorescence in decline index (Rfd). An increase in the content of chlorophyll-a (by 18%), b and carotenoids (by 7%) in the leaves after the pre-sowing treatment with I-Se has been shown. The content of chlorophyll-a in soybean leaves due to pre-sowing treatment with I-Se had the most significant increase after inoculation of P. agglomerans 9185 (20%). Due to the inoculation with a specific pathogen P. savastanoi pv. glycinea 9190 (after the pre-sowing treatment with I-Se), the content of chlorophyll-a tended to decrease, and the content of chlorophyll-b decreased by 7%. The carotenoid content in leaves increased significantly at inoculation of plant with facultative pathogens: P. agglomerans 9185 (by 28.2%) and P. savastanoi pv. syringae 8531 (by 20.7%). The chlorophyll a/b ratio increased in all experimental variants. Conclusions. 1. It was found that pre-sowing seed treatment with 1% I-Se chelated solution results in increasing disease resistance in nonspecific adult soybean plants, including increasing their thermotolerance. This is evidenced by increasing the peroxidase activity of leaves and the content of photosynthetic pigments in natural conditions: chlorophyll-a and carotenoids. 2. Inoculation of soybean plants with a specific pathogen causes an increase in catalase and peroxidase activities in leaves. The significant increase in peroxidase activity after plant inoculation with facultative pathogens was shown. 3. A significant increase in the content of chlorophyll-a in soybean leaves after inoculation with P. agglomerans 9185 and pre-sowing treatment with I-Se was observed. The significant reduction of chlorophyll-b in leaves due to the inoculation of plants with P. savastanoi pv. glycinea 9190 and pre-sowing treatment of the seeds with I-Se was shown. The most significant increase in the content of carotenoids in leaves was detected after inoculation of plants with facultative pathogens — P. agglomerans 9185 and P. syringae pv. syringae 8531. 4. Significant stimulation of both maximum quantum yield of PSII photochemistry (Fv /Fm) and the rate of carbon assimilation (Rfd) due to the pre-sowing seed treatment with I-Se and inoculation of plants with different strains of phytopathogens was observed. It resulted from the activation of antioxidant systems and increased need for assimilates due to changes in plant metabolism.

Ambiguous changes in photosynthetic parameters of Lemna minor L. after short-term exposure to naproxen and paracetamol: Can the risk be ignored?

Non-steroidal anti-inflammatory drugs (NSAID) are recently monitored in the aquatic environment. Naproxen (NPX), paracetamol (PCT) and their transformation products can influence the biochemical and physiological processes at the sub-cellular and cellular levels taking part in the growth and development of plants. This study aimed to compare the effects of NPX and PCT, drugs with different physico-chemical properties, on the growth and photosynthetic processes in Lemna minor during a short-term (7 days) exposure. Although duckweed took up more than five times higher amount of PCT as compared to NPX (275.88 µg/g dry weight to 43.22 µg/g when treated with 10 mg/L), only NPX limited the number of new plants by 9% and 26% under 1 and 10 mg/L, respectively, and increased their dry weight (by 18% under 10 mg/L) and leaf area per plant. A considerable (by 30%) drop in the content of photosynthetic pigments under 10 mg/L treatment by both drugs did not significantly affect the efficiency of the primary processes of photosynthesis. Values of induced chlorophyll fluorescence parameters (F0, FV/FM, ΦII, and NPQ) showed just a mild stimulation by PCT and a negative effect by NPX (by up to 10%), especially on the function of photosystem II and electron transport in both intact duckweed plants and isolated chloroplasts. Lowered efficiency of Hill reaction activity (by more than 10% under 0.1 – 10 mg/L treatments) in isolated chloroplasts suspension proved the only inhibition effect of PCT to primary photosynthetic processes. In intact plants, higher treatments (0.5 - 10 mg/L) by both NPX and PCT induced an increase in RuBisCO content. The results prove that the potential effect of various drugs on plants is hard to generalise.

Phytochrome-Dependent Regulation of Melon Resistance to Fusarium Wilt

The effect of pre-sowing seed treatment with light in the red spectral region on the resistance of melon plants (Cucumis melo) of the cultivar Kichkintoy to Fusarium wilt damage caused by Fusarium oxysporum f. sp. melonis was investigated. The directly-opposite effects of red and far red light on the degree of plant damage by the pathogen, which was determined by the special symptoms of the disease on the leaves and stems of plants, were revealed. When alternating seed treatment with red and far red light, the final effect was determined by the type of irradiation that acted last. The results of photobiological testing made it possible to establish the participation of the phytochrome system in the control of the resistance of melon plants of the cv. Kichkintoy to Fusarium wilt. It is shown that there is a high positive correlation between the parameters of chlorophyll fluorescence induction of leaves reflecting the functional activity of the photosynthetic apparatus and the degree of damage to plants grown from non-irradiated seeds and seeds irradiated with red light. The results of the conducted studies establish the possibility of effective regulation of the resistance of the melon cv. Kichkintoy to the defeat of F. oxysporum f. sp. melonis through photoactivation of the phytochrome system of seeds before sowing.
